The Mohawk Trail is a state highway, MA 2. I have driven it many times. The best time to drive it is in October during fall foliage, but it is popular with leaf peepers, so expect traffic. The worst time to drive it is during winter storms when snow and ice can slow the trip. From Greenfield to Williamstown (or reverse) could take a full day only if one spent a lot of time on hiking trails or other attractions along the route; otherwise, it is usually no longer than 2 hours. Berkshire County, MA has many inns and other fine places to stay, but tastes (and wallets) do vary.
